چکیده

By J. BURNS AND P. B. NEAME T lIE PERIODIC ACID-SCHIFF (PAS ) REACTION is generally accepted as the method of choice for demonstrating glycogen in blood cells. This reaction depends upon the presence, in glycogen, of vicinal glycols which are oxidized by periodic acid to dialdehydes. The latter are subsequently condensed with Schiff’s reagent to produce a magenta-red product, visible by light microscopy. Stoward1 has now conjugated acidified solutions of salicyloyl hydrazide witim dialdehydes formed from the periodic acid oxidation of vicinal glycols in guinea pig tissue sections. The reactive sites were said to fluoresce an intense blue on exposure to ultraviolet light. He recommended the use of salicyloyl hydrazide as the standard reagent for the demonstration of vicinal glycols in tissue sections. Because glycogen is a common constituent of certain normal and abnormal blood cells, and because its presence in some primitive cells has been used as a diagnostic aid in the differentiation of leukemias, we decided to determine if the periodic acid/salicyloyl hydrazide (PA-SH) method would be suitable for demonstrating glycogen in blood and l)one marrow cells.

Detecting glycogen in peripheral blood mononuclear cells with periodic acid schiff staining.

Periodic acid Schiff (PAS) staining is an immunohistochemical technique used on muscle biopsies and as a diagnostic tool for blood samples. Polysaccharides such as glycogen, glycoproteins, and glycolipids stain bright magenta making it easy to enumerate positive and negative cells within the tissue. In muscle cells PAS staining is used to determine the glycogen content in different types of mus...

Apoptotic and Necrotic Effects of Pectic Acid on Rat Pituitary GH3/B6 Tumor Cells

Background: Pectin is composed of complex polysaccharides that can inhibit cancer metastasis and proliferation with no evidence of toxicity. In the present study, the apoptotic and necrotic effects of pectic acid (PA) on the rat pituitary GH3/B6 tumor cells has been investigated. Methods: GH3/B6 cells were cultured in the Ham’s F12 medium enriched with 15% horse serum and 2.5% fetal bovine seru...

Comparison of Glycogen Positive Cells in Oral Smears with Random Blood Sugar Levels of Type 2 Diabetes Patients

Background: Even though much advancement in the medical field, diagnosis of diabetes mellitus (DM) needs an invasive procedure for drawing of blood and estimation of blood sugar levels. The purpose of this study was to analyse the glycogen content in exfoliated cells of oral mucosa as an adjunct in the diagnosis of diabetes mellitus. Aim: To assess the number of PAS (Periodic Acid Schiff) posit...

Investigation of amyloid deposition in uterine leiomyoma patients

Objects: To investigate the pathogenesis of amyloid presented in uterine leiomyoma. Methods: 36 uterine leiomyoma patients were recruited and divided into two groups according to Congo red staining results. 6 cases are Congo red staining-positive, and 30 cases Congo red staining-negative which represented amyloid positive and amyloid negative respectively. All patients’ serum total protein (TP)...

Identification of glycogen in thin sections of amphibian embryos.

Embryonic amphibian cells when examined with the electron microscope were observed to contain an abundance of small particles, approximately 325 A in diameter. The periodic acid/ Schiff reaction and enzymic digestion were employed to determine the nature of the particles, and from the results of these tests they were concluded to be glycogen. Treatment of thin sections with periodic acid/lead c...
